Discovery programs at a glance

Portrai’s discovery pipeline starts with Portrai ATLAS, our proprietary spatial biology database and data moat, and is translated through PortraiTARGET, our spatial transcriptomics intelligence engine for novel oncology target discovery. Each POR Series target is selected not only for expression, but for its spatial position within the tumor, selective enrichment against normal tissue, and biological fit with therapeutic modalities. From these targets, Portrai builds modality-fit asset candidates across ADC and RPT programs, while enabling future expansion into binder-based therapeutic modalities such as multispecific antibodies, TCEs, and ISACs. This allows Portrai to translate spatial cancer biology into a broader precision oncology therapeutic engine.

Target

POR001 is a Portrai ATLAS-derived novel stromal target designed to unlock the next generation of tumor-stroma-directed ADCs. Highly enriched in cancer-proximal stromal regions with limited normal-tissue expression, POR001 offers more tumor-restricted localization than conventional stromal targets such as FAP.

POR0011 is a fully human IgG antibody directed against POR001 and is being evaluated for development as a stromal-targeting ADC. POR0011 exhibits low-nanomolar binding affinity, high target specificity, target-specific cellular uptake, and robust internalization, supporting its potential for ADC applications across multiple solid tumor indications.

POR003 is a Portrai ATLAS-derived novel adenocarcinoma target selected for tumor-selective expression and ADC-compatible biology. Enriched in colorectal and pancreatic adenocarcinomas with limited normal-tissue expression, POR003 is positioned as a differentiated adenocarcinoma ADC target for tumors where deeper and more selective payload delivery remains a major unmet need.

POR0061 is a fully human IgG-based ADC candidate directed against POR006. With sub-nanomolar binding affinity, POR0061 is being developed as a SqCC-focused ADC candidate designed to leverage broad POR006 expression across squamous cancers.

POR0063Ab fragment–RPT Candidate

POR0063 is an antibody fragment–based RPT candidate directed against POR006. By combining antibody-fragment specificity with an RPT-compatible format, POR0063 is being developed as an additional precision radiopharmaceutical strategy for squamous cell carcinomas.
